Aine eesmärk on anda lai ülevaade analüütilise keemia instrumentide rakendamisest erinevate keemiliste omaduste ja protsesside (koostis, struktuur etc.) määramiseks. Tutvustatakse kaasaegse analüütilise keemia instrumentide põhialuseid, pakkudes teooriat süvitsi ning praktilisi oskusi.

This course is aimed to give a broad knowledge in the field of analytical instrumentation for the measurement of different chemical properties and processes (composition, structure, etc). The course covers fundamentals of modern analytical instrumentation, providing advanced background theory and practical skills.

Kursuse läbinud üliõpilane:
- mõistab kõrgemal tasemel mõningate kaasaegsete keemiliste analüüsinstrumentide ehitust ning tõõpõhimõtteid, spetsiifilist proovide lahutamist, optilist spektroskoopiat ning materjalide iseloomustamise tehnikaid;
- oskab hinnata erinevate analüüsimeetodite eeliseid ning puuduseid vastavalt proovi omadustele ning spetsiifilisetele analüütilistele probleemidele, suudab valida sobiva analüütilise meetodi väikestele molekulide, sünteetilistele polümeridele, enantiomeeridele ning biokeemilistele molekulidele valmistamiseks, lahutamiseks, detekteerimiseks ning kvantifitseerimiseks keerulistes maatriksites;
- hinnata analüütiliste lahutusmeetoditega saadud analüütiliste tulemuste ja valitud protseduuri kvaliteeti (analüüsimeetodi valideerimine);
- tuvastada ja lahendada peamisi ilmenvaid probleeme, mis tekivad analüüsi käigus, ning saada hakkama regulaarse instrumendi hooldusega

At the end of the course a student should be able:
- Understand the design and operating principles of some modern instruments at an advanced level, specifically separations, optical spectroscopy and material characterization techniques;
- evaluate strengths and limitations of different instrumental analysis techniques with respect to sample properties and to specific analytical problems and select a suitable analytical platform for the preparation, separation, detection and quantification of small molecules, synthetic polymers, enantiomers and biochemical analytes from complex matrices;
- judge the quality of obtained analytical results from analytical separation methods, and to evaluate the chosen operational procedures ( validation of chemical analysis methods);
- Identify the common problems arising during analysis and provide the potential solutions to this problems and to manage with regular maintenance.

Käesolev süvakursus sisaldab kaasaegsete füüsikaliste analüüsimeetodite (kromatograafia, spektroskoopia etc.) tõõpõhimõtteid ning praktilisi rakendusi proovide iseloomustamiseks. Uuritakse proovi erinevaid ettevalmistusi komponentide uurimiseks erinevatest maatriksitest, meetodite valideerimist ning tutvustatakse ka proovi lahutusprotsessi arvutil modelleerimist. Erilist rõhku pööratakse tööpõhimõtete tehnilisele arusaamisele, võimalike probleemide lahendamisele ja instrumendi hooldusele. Praktiline laboratoorne töö annab käelise kogemuse kvalitatiivses ning kvantitatiivses analüüsis erinevates teadustasemel meetodites (tahke faasi ekstraktsioon, superkriitiline ekstraktsioon, derivatiseerimine, gaasi- ning kõrgsurvevedelikukromatograafia, UV/Vis-spektromeetria, fluorestsents, massispektromeetria etc.)

This in-depth course covers the design, operational principles and practical application of modern physical methods (chromatography, spectroscopy, etc.) used in sample characterization. Sample preparation of analysis components from different sampling matrices/preparation forms, validation of chemical analysis methods and computer modelling of separation processes will be considered as well. The particular emphasis will be placed on „engineering “understanding of operation principles, troubleshooting and maintenance. Multi-week laboratory work provides hands-on experience in qualitative and quantitative analysis using a variety of research-quality methods (solid phase and supercritical fluid extraction, derivatization, gas and high pressure liquid chromatography, UV/Vis spectrometry, fluorescence, mass spectrometry etc).
