Tutvumine aminohapete, peptiidide ja valkude keemiliste omaduste ja struktuuriga. Anda ülevaade valkude struktuuri uurimise metoodikatest ning struktuurse proteoomika andmebaasidest. Omandada valkude visualiseerimise oskus.

Üliõpilased on omandanud põhiteadmised aminohapete ja valkude keemiliste omaduste ja valkude struktuurihierarhia kohta. Tunnevad valkude struktuuri uurimise metoodikaid, struktuurse proteoomika andmebaaside sisu ja ülesehitust ning on omandanud valkude visualiseerinise oskuse.

Students will get basic knowledge from chemical properties of amino acids, peptides and proteins, structural hierarchy of proteins, principles of structural methods, overview from protein structural databases and protein visualization.

Aminohapped - struktuur ja keemilised omadused. Peptiidside, struktuur, termodünaamiline ja kineetiline stabiilsus. Peptiidide keemiline süntees ja sekveneerimine. Peptiidahela konformatsioon. Dieedrilised nurgad, Ramachandrani kaart. Valkude kokkupakkimine (folding) ja denaturatsioon. Mittekovalentsete füüsikalis-keemiliste interaktsioonide roll valkude struktureerumises. Valkude struktureerumine in vivo - molekulaarsed šaperoonid, PDI, PI. Valkude posttranslatoorne ja keemiline modifitseerimine. Valkude glükosüleerimine. Meetodid biopolümeeride ruumilise struktuuri määramiseks: röntgenkiirte difraktsioon, NMR, elektronmikroskoopia. Valkude struktuuri hierarhia, supersekundaarstruktuur ja domeenide struktuurne klassifikatsioon. Valkude struktuuri dünaamika. Transkriptsioonifaktorite struktuur ja toimemehhanismid. Valk_valk ja valk-nukleiinhape interaktsioonid, molekulaarse äratundmise põhimõtted. Membraanvalkude struktuuri uurimise metoodikad. Oluliste membraanvalkude struktuur ja funktsioneerimine. Valgustruktuuride visualiseerimine ja võrdlemine.

Standard amino acids in proteins: nomenclature, structure of side chains and chemical properties. Chemical and biochemical modification of amino acids. Structural levels in proteins. Peptide bond, thermodynamic and kinetic stability. Conformation of peptides, dihedral angles, Ramachandran plot. Folding of proteins in vitro and in vivo. Molecular chaperones, PDI, PI. Methods for structural characterizatrion of proteins: X-ray fifractometry, 3D-NMR, electron microscopy. Structural hierarchy of proteins, secondary, supersecondary, domain, tertiary and quaternary structure, supramolecular complexes. Classification of structural domains. Structural dynamics in proteins. Structure of transcription factors. Protein-protein, protein – nucleic acid interactions, principles of molecular recognition. Membrane proteins – methods for their structural studies and mechanisms of functioning of major membrane protein complexes. Visualization and comparison of protein structures.
